    摘要:

    听力障碍中约60%由基因缺陷所致,称为遗传性耳聋。遗传性耳聋的基因治疗虽取得进展,但尚未实现有效防控。遗传性耳聋通常是由内耳细胞受损造成。诱导多能干细胞(iPSC)可以从体细胞诱导分化为多能干细胞,进而通过二维或三维诱导等方式在体外诱导为毛细胞、螺旋神经元等内耳感觉上皮细胞的类细胞或类器官,受到广泛关注。随着iPSC技术的不断进步,其在遗传性耳聋研究领域的应用变得日益广泛。本文将从iPSC的优势、诱导策略、应用方式等方面对iPSC在遗传性耳聋研究中应用的进展进行综述。

    Abstract:

    Approximately 60 % of hearing impairment is caused by genetic defects, known as hereditary hearing loss. Although gene therapy for hereditary hearing loss has made progress, effective prevention and control have not yet been achieved. Hereditary hearing loss is usually caused by damage to the inner ear cells.Induced pluripotent stem cells (iPSCs) can be derived from adult cells and induced to differentiate into pluripotent stem cells. Subsequently, through two-dimensional or three-dimensional induction, they can be further induced in vitro into inner ear sensory epithelial cells like cells, such as hair cells, spiral ganglion neurons and organoids. In recent years, with the development of iPSC technology, its application in the research of hereditary hearing loss has become increasingly prominent. This review will provide an overview of the progress in the application of iPSCs in the research of hereditary hearing loss, covering aspects such as the advantages of iPSCs, induction strategies, and application methods.
