    摘要:

     目的通过对比鼻咽癌细胞610B和HNE2的放疗敏感性,为临床鼻咽癌不同分期放射治疗提供依据。方法鼻咽癌细胞6-10B和HNE2用于放疗敏感性实验检测,通过集落形成实验检测细胞对放疗的敏感程度、流式细胞术检测细胞周期、流式细胞术与Western blot检测细胞凋亡、免疫荧光结合Westen blot检测DNA损伤标志物γH2AX表达水平。结果集落形成实验结果初步看出HNE2(IC50约3.5 Gy)对放疗的敏感性强于6-10B(IC50约5 Gy);细胞周期结果可以发现放疗后,两株细胞均出现G2M期周期阻滞;HNE2细胞放疗引起的DNA损伤和凋亡水平均高于6-10B。结论鼻咽癌细胞HNE2对放疗的敏感性强于6-10B,为鼻咽癌的不同分期治疗提供临床参考。

    Abstract:

     ObjectiveTo provide basis for radiotherapy of nasopharyngeal carcinoma (NPC) in different stages via comparing the radiosensitivity of NPC cells 610B and HNE2.MethodsNPC cells 610B and HNE2 were used for radiosensitivity test. Their sensitivities to radiotherapy were detected by colony formation test. Cell cycle was monitored by flow cytometry. Apoptosis in different groups was inspected by flow cytometry and Western blot. Immunofluorescence combined with Western blot was used to measure the phosphorylated histone γH2AX reflecting the level of doublestrand DNA damage induced by radiotherapy.ResultsThe results of colony forming experiment showed that HNE2 (IC50≈3.5 Gy) was more sensitive to radiotherapy than 610B (IC50≈5 Gy). The results of cell cycle showed that both cells showed G2Mphase cycle arrest after radiotherapy. DNA damage and cell apoptosis.ConclusionHNE2 is more sensitive to radiotherapy than 610B, which provides reference for the treatment of NPC at different stages.
